General Description Of Hirudin
Hirudin is a highly effective and very specific anticoagulant (Direct Thrombin Inhibitor) which specifically inhibits formation of fibrin and activation of factors V, VIII and XIII by forming irreversible, noncovalent 1:1 complexes with a-thrombin. Hirudin is a 65 amino acid polypeptide, 7 kDa protein naturally originating from the Hirudinaria manillensis, the Malaysian local buffalo leeches. It is the most potent thrombin inhibitor known to date.
Clinical Relevance
Must be administered intravenously or subcutaneous injection.
Predominantly cleared by kidney, it should not be used in patient with renal insufficiency.
Plasma half-life of approximately 60 minutes after intravenous administration and 120 minutes after subcutaneous injection.
Monitored using the activated partial thromboplastin time (aPTT).
